PENENTUAN KADAR TOTAL FLAVONOID DAN NILAI SPF (SUN PROTECTION FACTOR) EKSTRAK ETANOL RIMPANG TEMU IRENG (Curcuma aeruginosa Roxb.) SECARA SPEKTROFOTOMETRI UV-VIS
Abstract
Temu ireng contains phenolic compounds in the form of flavonoids. This study aims to determine the total flavonoid content and SPF value of the ethanol extract of Temu Ireng (Curcuma aeruginosa Roxb.) Ethanol extract. Extraction was carried out using the maceration method with 96% ethanol solvent, then a qualitative test of flavonoids with the addition of concentrated Mg and HCl metals and a quantitative test with running quercetin and making a quercetin standard solution as a comparison with a concentration of 20 ppm, 30 ppm, 40 ppm, 50 ppm, 60 ppm and Temu ireng ethanol extract test solution with a concentration of 1500 ppm and 50,100,150,200 ppm. The absorption was measured with a UV-Vis spectrophotometer at a wavelength of 376 nm. Then the SPF test was carried out by making a test solution with a concentration of 50 ppm, 100 ppm, 150 ppm, 200 ppm. The absorption was measured with a UV-Vis spectrophotometer at a wavelength of 290-320 nm with 5 nm intervals. The results showed that the total flavonoid content at a concentration of 1500 ppm was 120.53% while at a concentration of 50,100,150 and 200 ppm were 4.58%; 8%; 10.83%; 13.78%. The SPF value of the Temu ireng rhizome extract with a concentration of 50 ppm, 100 ppm, 150 ppm, and 200 ppm, respectively, were 1.067 (not included in the category); 2,311 (minimal protection); 3.47 (minimal protection); and 4,541 (moderate protection).
Temu ireng mengandung senyawa fenolik berupa flavonoid. Penelitian ini bertujuan untuk mengetahui total kandungan flavonoid dan nilai SPF ekstrak etanol Rimpang Temu Ireng (Curcuma aeruginosa Roxb.). Ekstraksi dilakukan menggunakan metode maserasi dengan pelarut etanol 96%, kemudian dilakukan uji kualitatif flavonoid dengan penambahan logam Mg dan HCl pekat dan uji kuantitatif dengan running kuersetin dan pembuatan larutan baku kuersetin sebagai pembanding dengan konsentrasi 20 ppm, 30 ppm, 40 ppm, 50 ppm, 60 ppm dan larutan uji ekstrak etanol Temu ireng dengan konsentrasi 1500 ppm dan 50,100,150,200 ppm. Diukur serapannya dengan Spektrofotometer UV-Vis pada panjang gelombang 376 nm. Kemudian dilakukan uji SPF dengan pembuatan larutan uji dengan konsentrasi 50 ppm, 100 ppm, 150 ppm, 200 ppm. Diukur serapannya dengan spektrofotometer UV-Vis pada panjang gelombang 290-320 nm dengan interval 5 nm. Hasil penelitian menunjukkan kandungan total flavonoid pada konsentrasi 1500 ppm sebesar 120,53% sedangkan pada konsentrasi 50,100,150 dan 200 ppm berturut-turut yaitu 4,58%; 8%; 10,83%; 13,78%. Nilai SPF ekstrak etanol rimpang Temu ireng dengan konsentrasi 50 ppm, 100 ppm, 150 ppm, dan 200 ppm, berturut-turut yaitu 1,067 (tidak masuk kategori); 2,311 (proteksi minimal); 3,47 (proteksi minimal) ; dan 4,541 (proteksi sedang).
